Actually, Toriyama made sure that the change is apparent in the manga, recognizing a SSJ2 is very easy (and Majin Vegeta was one, I think). It's the anime where it's more ambiguous.

After reading the manga I was sure that it was kind of a warm-up, Buu was slowly, ostentatiously building up the momentum of the punch just to show he can take his sweet time when fighting Vegeta. I'm not sure about the anime, don't remember the episode.

The screenshot seems to indicate that the transformations are chosen in a way similar to costumes in Budokai games, and they don't count as multiple characters.
