Does it matter what region the DVD's in for it to be ripped by these programs?

The programs can read it so long as your DVD drive can read it. I know that DVD Decrypter has an option to change the drive's region, but it does so in the same way that you'd change it yourself through Windows or whatever OS you use. And if you do want to change it, remember that many drives can only have their region changed five times before the region becomes locked.

Are there any that will only change once before locking?

I'm not sure, but I think your system tells you how many changes you have left when you attempt to change it.

DVD Decrypter will brute-force the rip (usually at a lower speed) even if the region doesn't match your drive. I typically rip and re-burn R2 and R3 movies that I purchase, and watch them on my standard R1 DVD player this way.
