Black being skinner, tanner and having less shading is just an out of universe artistic choice to have something to differentiate them by, just like how Future Trunks has blue hair, and regular Trunks has purple hair.

Goku's body has the same DNA, but Goku isn't simply Goku's body, and Goku Black isn't simply Goku's body. The key difference between them is the spirit.

Biology Student here,

In terms of genome, Goku and Goku Black would be the same. The genome is the overall set of all of your body's genes. In twin brothers, for example, the overall sequence of genes is exactly the same, but variations can occur.

The most common ones are long-term mutations, caused by different life experiences, different personalities and what-not. This results in one of the teens, being, for example, skinnier than the other, because stress can be a skinning agent for some people. I just wanted to point this out because genetic coding isn't the only thing that makes people... people. Life experience dictates which genes get activated and which get deactivated, making even the most identical twins diverge.

Now for Goku and Goku Black:

Right from the moment that Zamasu asked to change bodies with Goku, I imagine that their DNA would be exactly the same. But that's just about it.
From the moment of now Goku Black's newly acquired body to the point of his appearance, a lot of things happened. In that time-frame, Goku Black's emotions and overall personality are so different than Goku's, that I don't find it difficult to believe that the body started changing...

Just imagine:
- Goku Black has a plan to carry out. He made himself have goals, deadlines, and all of that generates one thing: stress.
- Goku Black lifed all of his life as a Kaio, and because of that he has that almost like 'divine' posture that all the Kaioshin have, and so having that posture can change one's vertebral chord. I imagine that's why Goku Black always looks kind of taller than Goku even though they have exactly the same height: Posture.
- Emotions play a big part in hormone regulation: Goku Black's hate for mortals and his sense of duty regarding the Ningen Zero Keikaku, can have retaliations on his body's health: Firstly, the first reaction one's body has when that certain individual is feeling angry is to produce adrenaline, which makes the heart rate go up, resulting in a lot higher fat burn. That, mixed with the stress, could explain Goku Black's thinnier figure.
Then, all of Goku Black's genome begins functioning towards addapting to those emotions as a permanent state, whereas in Goku those emotions were triggered only when something really bad happened.

Now, in conclusion, gene activation and repression, alongside with different hormone regulation would be enough to state that Goku and Goku Black's DNA is different. From the moment that Zamasu acquired Goku's body, the 2 individuals life experiences diverged so much that, in one side, we have a stable life, with friendship, love, being close with people, and also a commitment to get stronger, which is always healthy, wheres on the other side we have anger, hate, an evil determination, anti-social behaviour and overall negative feelings. Just plain and simple if you take over someones body or swap bodies with them, the DNA remains the same. It is still their body and everything of theirs. Think of it as nothing more than a mind-swap.

Well, I provided a well based explanation on why I think the DNA wouldn't be the same, if you want to consider it or not, it's up to you.

Youre thinking too real world in the last part maybe. In this fictional universe we know with 100% fact the spirit and body are two separate entities.

So while their body is the same at a DNA structure level they are entirely different people, so his body may be related to his children but overall as a full entity id say no.

Its interesting to ponder but real world biology doesnt apply to DB for an incredible number of reasons.

Well, I think if they did DNA tests, Goku Black would still reveal himself as in the 'Son' family, cuz what's changed in their genome is the "hormone" and some appearance genes that got rearranjed..
