Vaccine Man is clearly a reference to Piccolo. In english, german and mexican spanish dubs, both characters share the same VA's.
In the japanese original version, Vaccine Man is voiced by Ryusei Nakao, the original voice of Frieza, and in the brazilian portuguese dub, Vaccine Man is voiced by Wellington Lima, the brazilian voice of Majin Boo.

Witch is clearly a reference to Vegeta at 1:14.

I disagree. Vaccine Man is based on Baikinman, an antagonist from Anpanman, which is what One Punch Man was originally intended to be a a parody of.

It has similarities with him, as the voice as well (Ryusei Nakao).
